Is it legal to operate an express company on a business visa in Thailand?

You won't be granted the visa for such a set up, you need a million documents to secure a non b visa and you won't pass any of the terms.

Do it online then it doesn't matter. Takes a few minutes at most

Where can I buy Methotrexate in Thailand without a prescription?

Prescription only. It is very cheap, I wouldn't mess around trying to find a drug like that without prescription. As mentioned above you need to be monitored especially your liver.
