What are the requirements for extending my tourist visa in Thailand regarding TM30 registration?

I entered with a 60 day tourist visa (US national). Did an extension in late September just before the amnesty ended. Went back to close the loop in mid October and was given one extra month. The extension now ends on November 30.

I'm going for another extension but I've heard that TM30 is now required? I've always stayed at a friend's (owner's) place and never had to have TM30 done for any extension before so there is no signed agreement or anything.

Is the method here still valid even if the TM30 has never been filed before?

Yes. I just extended this morning and was asked to show my proof of TM30 registration. There are stories that some were not asked, but if you are unlucky to get asked, you will have wasted a trip as they won't proceed without it.
