I have a category O non-immigration type Visa that has been extended until May 8, 2020. However, my work permit will expire on February 16th, 2020. If my work permit expires, will the extension of my Visa automatically be voided, and I will need to leave the country?

You're NOT on any visa, you're on an extension and when the reason for the extension changes (like your work permit expires) you need to go and cancel your extension effective that date
IF you're not gonna go renew your work permit, then when it runs out you need to go turn it in at the Ministry of Labor and then go immediately and cancel your extension of stay. Then you either leave that day OR you buy the 7 day extension denied stamp that gives you a week to get out of thailand for 1900baht
