TLDR : Answer Summary
Applying for a Non-B visa in Thailand while holding a work permit depends largely on your job and your employer's ability to provide the necessary documentation for immigration. The process is generally less complicated if you apply from a consulate overseas. It is advised to involve your employer in discussions with immigration to determine feasibility.

In country conversion to a Non B is completely dependent on the job you will be doing and the company set up. If it can provide the documentation that immigration needs then it can be done.

Far less onerous documentation to get a non B from a consulate overseas.

You’d probably need to take your employer to immigration and have them work out if it’s possible.
