I am on a non-imm O extension. I know to get a reentry stamp on the way out. When I return do I need to go to my local immigration office, or does my 90 days automatically reset from my arrival?

TLDR : Answer Summary
When returning to Thailand on a non-immigrant O extension, your 90-day reporting starts again upon reentry. You need to calculate the days yourself for the first 90-day report after returning. Additionally, it's required to report your address at the immigration office within 24 hours of your return and file a Tm30 form.

As John said you must file a Tm30 at immigration

Many immigration offices also require you to report your address within 24 hours of returning from abroad

how is this enforced?

it's enforced when you go to get your next extension and they see that the TM30 filing date is NOT sync'd with your most recent arrival date. (that's when they usually tell you go to file a NEW TM30 and come back)

so they will punish you by making you wait in another line?

At least they donโ€™t make you wash their car or sit in the corner facing the wall with a silly hat on.

they aren't punishing you at all, they're making you follow the rules is all ๐Ÿ˜‰

It is YOUR responsibility to make sure you or your land lord file a TM30 when you come back into the country (some offices even make you do it when you come back from domestic travel too ๐Ÿ˜ฎ )

I mean it is what it is, and it's been a rule for a LONG time (20+ years I believe) but it's only the last few years where it's been enforced.

43 years. The Immigration Act of 1979. Section 38.

it's not enforced per se. But next time you need to interact with immigration you can be fined. To be more correct its the owner of the property that should be fined but it invariably becomes the foreigners responsibility

your 90 days, which is a report that you're 90 days continiously inside Thailand, it stops at the moment you leave Thailand and it starts at the moment you enter Thailand. You need to calculate the days by yourself to make the first 90 day report.
