I had 11 days overstay in September which I've already paid for the fine. Now I'm on my second covid extension. Is it possible to apply for work visa and work permit at this point? I'm currently on RTV.

IF I was to guess I'd say NO, if you ever went on an overstay during this current time in thailand you can't get it done. BUT the immigration office you deal with is who you need to talk to and show your passport to

I've yet to ask them. What I've heard is that if there's an "overstay" on my passport, the only way to change to Non-O, Non-B or any type of visa is to leave the country and come back in.

You need to talk to your local immigration office about that. If it's Bangkok, you cannot apply for any in-country visa when you are on an overstay or there is the period you were on overstay since your last entry into Thailand.
The basic rule is clear.
"The application must be submit more than 15 days before visa expiration and, in case of overstaying in Thailand, application could not be submitted."
It depends on the local immigration office how they treat the case that you paid an overstay fine and keep staying on some type of extension.
My business partner had a 2 day overstay about 4 months ago. He’s currently attempting to switch from a Covid extension to a Non-B. The immigration officer didn’t even look at his paperwork after seeing the overstay in his passport. Told him he has to leave and come back in on the Non-B. Open and shut case for him unfortunately. But, that’s not saying it can’t or won’t be different for you…just probably best not to get your hopes up.
If you can go from a 60 day extension of stay based on unable to travel due to COVID-19 to an in-country Non-B visa based on employment will depend on you, the employer, and the immigration officer. Have you asked your employer if it's possible?
