No, you're visa gets valid at the moment it is attached into your passport and as this will be a Single Entry Non Immigrant B visa get stamped used and invalid as soon as you cross the border into Thailand. The Work Permit gets valid at the moment it is stamped and signed by the Ministry of Labor, your contract gets valid when signed by you and your employer. BUT you can only start to work if the visa is in your passport and you got the Admitted to Stay based on that visa and the work permit is in your pocket.
