I doubt so . If visa was firstly done in Samui they will make you go Samui to get your extension. Speaking from experience I was made to go back to Phuket for the process
are you talking about "closing out an under consideration" stamp (where you go back to who issued the original stamp) OR are you talking about applying for a new 60 day covid extension?
Most offices as long as you can prove you stay in the area they cover will let you file a covid extension with them..

you might have to prove you actually live in the area where you're applying for the extension (as in you or your land lord filed a TM30 notification of foreigner in residence) but there isn't a problem getting a 60 day covid extension at either immigration office in Surat Thani province

If you are tourist(non-resident), easiest is check into a Bangkok hotel a few days before you go to Bangkok immigration. Ask hotel to give you a copy of their TM30 registration for you. Bring it with you, and a copy of your hotel booking, when you go to immigration to apply for the extension.
