Can I apply for a non o multiple entry when I arrive in thailand or do I need to apply before arriving?

You can apply for a re-entry permit before leaving Thailand, allowing you to keep the stamp alive but not adding extra time.
If you're looking for the 1 year multiple entry non-O visa that gives you 90 days each time you enter Thailand, that's only available for married to Thai and only available outside of Thailand.
then you can't do it. Marriage non-O usually gets a 30 day under consideration stamp and they might want to do a home visit. 3 weeks is not enough time. You'll need to do it when you return or you can get it from Thai embassy in your home country and then apply for a re-entry permit before you leave for work, then apply for your 1 year extension when you return from your job.
If you already have the 400,000 in your Thai bank account shouldn't be any issues in applying for your extension.

Also if I come in on visa exempt for 3 weeks then return 26 days later will they let me enter to change too the non o spouse visa ,?
