After the non-o (Thai wife) yearly extension got approved, does the 400,000 still need to be left in the account for the whole year until the next extension?

No, only during the 2 months prior application and at request of the Immigration Office during the 1 month under consideration. Rest of the year you can spend it all.
I have provided them with a 6 month statement. The IO now wants 1 year. I was told my visa will not pass because I removed money after getting my first extension, but I could receive the extension if ...(I don’t think I can post this online). There was constant mention of nationality in a demeaning way throughout the whole conversation.
