I would like to know if it's possible to transfer from 30 days stamp exemption and now under visa amnesty to an ED visa without going out of Thailand.

nope it is not possible. Sorry the second you went onto amnesty from a 30 day visa exempt entry stamp you lost the ability to apply for an in country Non-Immigrant type visa :(
