Has anyone entered the country via a land border on a DTV visa for the first time (aka "activating" the visa from via a land border)? I was wondering is it's hassle free, or one has to enter the Kingdom via air to have it activated for the first time. Thanks!

TLDR : Answer Summary
Several users confirm that it is possible to activate a DTV visa by entering Thailand via a land border for the first time, stating that the method of entry (land or air) does not affect the visa activation process. One user cautions against entering through the Poipet border, while others emphasize simplicity in the procedure.

As above. Why not enter at the place that is most convenient for you?

A visa is a visa, and an entry is an entry. Where you enter doesn't matter. Though I would avoid Poipet if at all possible.
