TLDR : Answer Summary
Yes, US passport holders need a visa to enter Cambodia when traveling from Thailand. They can obtain a visa on arrival for $30 or apply for an E-visa online beforehand. It's important to have cash in US dollars, as other currencies, including Thai baht, are generally not accepted upon arrival.

Most nationalities require a visa to enter Cambodia; some are allowed to buy an e-visa online, some must apply at an embassy before travel, some are allowed to apply for a visa on arrival. You have to look it up to see what's required/allowed for the passport you carry.

The following nationalities do NOT require a visa to enter Cambodia:

Brunei

Indonesia

Laos

Malaysia

Maldives

Myanmar

Philippines

Seychelles

Singapore

Thailand

Vietnam

If you’re flying to Phnom Penh you can fill out the form at immigration, it’s done on a tablet, $30USD fee, be sure to have that amount in cash, Thai baht is not accepted.
