TLDR : Answer Summary
Yes, US passport holders need a visa to enter Cambodia when traveling from Thailand. They can obtain a visa on arrival for $30 or apply for an E-visa online beforehand. It's important to have cash in US dollars, as other currencies, including Thai baht, are generally not accepted upon arrival.
What difference does it make from where you coming to Cambodia? You need visa fro wherever you arrive

Yes, I’m U.S. citizen and went to Cambodia 4 years ago. The visa on arrival at Phnom Penh airport ,not sure if visa on arrival at the border(if by land). Best bet is e-visa.
Most nationalities require a visa to enter Cambodia; some are allowed to buy an e-visa online, some must apply at an embassy before travel, some are allowed to apply for a visa on arrival. You have to look it up to see what's required/allowed for the passport you carry.
The following nationalities do NOT require a visa to enter Cambodia:
Brunei
Indonesia
Laos
Malaysia
Maldives
Myanmar
Philippines
Seychelles
Singapore
Thailand
Vietnam

If you’re flying to Phnom Penh you can fill out the form at immigration, it’s done on a tablet, $30USD fee, be sure to have that amount in cash, Thai baht is not accepted.
